clonage, PCR-RFLP | Index. décimale : | P342-Bactériologie et microbiologie du sol : azote dans le sol; inoculation, rhizobium, micorhyse | Résumé : | Pour étudier l’influence de différents modes de gestion des terres sur la diversité et l’activité des bactéries du sol, des échantillonnages de sol sont réalisés sur 4 parcelles d’un essai au champ situe à Thyssé Kaymor (Sénégal) : jachère naturelle, jachère améliorée à Andropogon gayanus, jachère améliorée à Acacia holosericea, sol sous culture continue (rotation mil - arachide). Les prélèvements sont effectués selon la chronoséquence suivante : à la défriche des jachères âgées de 4 ans, à la récolte de la première culture (3 mois après la défriche) et enfin
après la sixième culture (6 ans après la défriche). Un fractionnement physique du sol est réalisé pour isoler les habitats microbiens en fonction de leur granulométrie. Des mesures chimiques (carbone et azote organiques, dénitrification potentielle) et des analyses moléculaires (PCR-DGGE du gène 16S ADNr, clonage-RFLP du gène narG) sont effectuées sur les habitats >2000 μm (macro-agrégat) et 2-50 μm (microagrégat).
Les résultats obtenus montrent que la communauté microbienne hébergée dans l’habitat 2-50 μm est caractérisée par une activité potentielle et une diversité génétique plus élevées que la communauté présente dans les agrégats >2000 μm. La comparaison des différentes jachères indique que la présence d’Andropogon gayanus stimule l’activité et la diversité des communautés microbiennes par rapport à la jachère à Acacia holosericea.
L’analyse des profils RFLP narG de la communauté microbienne dénitrifiante présente dans l’habitat 2-50 μm indique que la diversité de ces communautés est liée au type de végétation et à l’antécédent cultural. |

This study assessed the effect of two representative low and high quality residues (Zea mays and Crotalaria retusa respectively) on the formation and stability of soil aggregates. The formation and stability of aggregates, soil biomass, root biomass, fungal hyphae length, C mineralization and chitinase activity, as a specific biomarker of the activity of fungal populations, were measured under controlled conditions over 120 days. In both the
control and amended soils, there were more macroaggregates (>2000 mm) and mesoaggregates (250 e2000 mm) than microaggregates (50e250 mm and <50 mm). The formation of macroaggregates and stability (MWD) were not significantly affected by the quality of residues. Amendment with organic residues shifted the distribution of the aggregate fractions. The macroaggregates increased by 26% with crotalaria and by 35% with maize residues while mesoaggregates decreased by 18% with crotalaria and by 26% with maize residues and microaggregates decreased by 8% with crotalaria and by 9% with maize residues. This study also onfirmed that macroaggregates are formed from micro- and mesoaggregates. The total microbial biomass was significantly higher in soil amended with maize residues compared to oil with crotalaria residues and the control soil although the fungal hyphae length decreased when the soil was amended with either crop residue. Chitinase activity is the most pertinent indicator associated with acroaggregation stability. Adding mineral N (equivalent to 120 kg N ha1 as urea) to the residue increased microbial biomass and reduced fungal hyphae length but had no effect on macroaggregate formation and fungal activity. These observations suggested that, for short term incubation of soil amended with residues, fungal activity plays a greater role in aggregation in sandy soils than the fungal population density. |

The presence of shrub roots and litter inputs should have implications for soil nutrient pool sizes but there is limited information on the interactions of these shrubs with microbial communities involved in biogeochemical processes. Therefore, the objective of this study was to determine the microbial composition and functional capacity of soil from the rooting zone of pearl millet (Pennisetum glaucum) grown in the presence or absence of the shrub Piliostigma reticulatum in Senegal. Soil samples were collected from a long-term field study where millet was cultivated alone or intercropped with P. reticulatum with annual incorporation of coppiced shrub residues. Higher nutrient contents and distinct differences in microbial communities (DGGE profiles) were found between soils from beneath the canopy compared to soil outside the influence of shrubs. The catabolic response profile (MicroResp™) showed that the soil microbial community at both shrub and non-shrub sampling locations, metabolized a wide range of substrates. Trehalose that can work as a signaling molecule was more rapidly degraded in the rooting zone of millet growing in the presence of P. reticulatum over millet alone. Urease, arylsulfatase and dehydrogenase activities in the millet root zone soil were higher when intercropped with P. reticulatum which indicates enhanced potential of biogeochemical processes to proceed in the presence of this shrub. It is concluded that the native shrub P. reticulatum promotes a more diverse and active microbial community in the rooting zone of millet and further indicates greater potential to
perform decomposition and mineralize nutrients. |

Annual application of manure led to the highest soil microbial biomass and enzyme activities. Investigations indicated that only microbial biomass and β-glucosidase activities were affected during the cropping season. Phosphatase and FDA enzyme activities did not depend on the crop development stages. The application of N fertilizer modified phosphatase and FDA enzyme activities, the activities being higher in soils amended with N fertilizer. The bacterial community structure was analyzed by PCR-denaturing gradient gel electrophoresis (PCR-DGGE) targeting the eubacterial 16S rRNA gene. Cluster analysis of PCR-DGGE patterns showed two major clusters, the first containing the mineral fertilization and straw treatments and the second, the straw + urea, manure and manure + urea treatments. Sorghum grain yields were the highest for
manure treatments. In this long-term experiment, applying straw did not produce a better grain yield than that obtained in the un-amended plot. |
