clonage, PCR-RFLPIndex. décimale : P342-Bactériologie et microbiologie du sol : azote dans le sol; inoculation, rhizobium, micorhyse Résumé : Pour étudier l’influence de différents modes de gestion des terres sur la diversité et l’activité des bactéries du sol, des échantillonnages de sol sont réalisés sur 4 parcelles d’un essai au champ situe à Thyssé Kaymor (Sénégal) : jachère naturelle, jachère améliorée à Andropogon gayanus, jachère améliorée à Acacia holosericea, sol sous culture continue (rotation mil - arachide). Les prélèvements sont effectués selon la chronoséquence suivante : à la défriche des jachères âgées de 4 ans, à la récolte de la première culture (3 mois après la défriche) et enfin
après la sixième culture (6 ans après la défriche). Un fractionnement physique du sol est réalisé pour isoler les habitats microbiens en fonction de leur granulométrie. Des mesures chimiques (carbone et azote organiques, dénitrification potentielle) et des analyses moléculaires (PCR-DGGE du gène 16S ADNr, clonage-RFLP du gène narG) sont effectuées sur les habitats >2000 μm (macro-agrégat) et 2-50 μm (microagrégat).
Les résultats obtenus montrent que la communauté microbienne hébergée dans l’habitat 2-50 μm est caractérisée par une activité potentielle et une diversité génétique plus élevées que la communauté présente dans les agrégats >2000 μm. La comparaison des différentes jachères indique que la présence d’Andropogon gayanus stimule l’activité et la diversité des communautés microbiennes par rapport à la jachère à Acacia holosericea.
L’analyse des profils RFLP narG de la communauté microbienne dénitrifiante présente dans l’habitat 2-50 μm indique que la diversité de ces communautés est liée au type de végétation et à l’antécédent cultural.Exemplaires

Réponse des plantes Résumé : Casamance is a region in southern Senegal that traditionally produces rainfed rice, but Senegal produces only 1/3 of its rice consumption. Lowland areas, where rice is primarily produced, have acid sulfate soils with low pH and potential aluminum and iron toxicity. The goal of this work was to determine if soil amendments can alleviate soil acidity, counteract the negative biogeochemical effects that occur in flooded conditions, and increase rice yield. A two-year experiment was conducted to test the following soil treatments – agricultural lime, pulverized oyster shell, biochar, and control (no amendment) – in flat and raised beds. Plots amended with lime and shell materials had increased soil pH, base saturation, Ca, and cation exchange apacity. Meanwhile, biochar elevated particulate organic matter and C:N ratios. Exchangeable Fe and Al were negatively correlated with soil pH, while Geobacteraceae populations (Fe reducing bacteria) increased with pH. A greater proportion of the total Fe was strongly bound in fractions that were less bioavailable in plots amended with shell or lime, and overall rice yields were significantly higher following amendment with shell or lime. During the second growing year these effects iminished, suggesting that liming effects did not persist as expected. These results demonstrate the benefits of soil amendments that raise soil pH and suggest that this effect operates by influencing overall soil nutrient availability to rice plants, but further research is needed regarding the timing and sustainability of the beneficial liming effect. We studied the impact of different fertilizer regimens in an agricultural long-term observatory in Burkina Faso at three different plant development stages (early leaf development, flowering, and senescence) of sorghum cultivars. Using real-time PCR, we investigated functional microbial communities involved in key processes of the nitrogen cycle (nitrogen fixation, ammonia oxidation, and denitrification) in the rhizosphere. The results indicate that fertilizer treatments and plant development stages combined with environmental factors affected the abundance of the targeted functional genes in the rhizosphere. While nitrogen-fixing populations dominated the investigated communities when organic fertilizers (manure and straw) were applied, their numbers were comparatively reduced in urea-treated plots. In contrast, ammonia-oxidizing bacteria (AOB) increased not only in absolute numbers but also in relation to the other bacterial groups investigated in the urea-amended plots. Ammonia-oxidizing archaea exhibited higher numbers compared to AOB independent of fertilizer application. Similarly, denitrifiers were also more abundant in the urea-treated plots. Our data imply as well that, more than in moderate regions, water availability might shape microbial communities in the rhizosphere, since low gene abundance data were obtained for all tested genes at the flowering stage, when water availability was very limited. la production agricole est devenu une priorité pour les acteurs du développement. L’article porte sur l’estimation précoce de la
production cotonnière avant la récolte au Sénégal, par voie d’expertise de la SODEFITEX. Objectifs. L’étude a pour objectifs d’expliciter les connaissances mobilisées par les experts pour prédire le rendement, eu égard à l’incertitude sur les conditions climatiques à venir, et d’évaluer la performance des prévisions, selon le niveau territorial considéré. Méthode. Pour atteindre ces deux objectifs, l’article s’appuie sur l’analyse statistique de la production cotonnière au Sénégal
de 2004 à 2015, combinée à une enquête réalisée auprès de 32 experts de la SODEFITEX. Résultats. L’analyse des résultats d’enquête met en évidence une prévision principalement établie sur la base de l’état des cultures, des pratiques culturales au premier rang desquelles la date de semis et, pour certains experts, de l’historique de la production et son environnement organisationnel et social. La comparaison des prévisions avec la production effective montre que les experts surestiment systématiquement les niveaux de production inférieurs à 1 000 kg.ha-1, en conditions climatiques défavorables. Conclusions. Dans un contexte de faible documentation scientifique sur l’estimation précoce de la production par voie d’expertise, les résultats de l’étude éclairent sur les connaissances mobilisées et la performance des prévisions délivrées, ouvrant ainsi la voie à une critique rationnelle de l’expertise. La diversité de points de vue mise en évidence par l’analyse multicritère, entre niveaux hiérarchiques et entre agents de même niveau, souligne le besoin de confrontation des connaissances pour faire émerger les variables explicatives nécessaires à la prévision et tendre vers une objectivité accrue de l’expertise face aux aléas climatiques récurrents.Exemplaires

Décrite comme une grande force de l’évolution, elle est causée par divers facteurs génétiques ou physiologiques, influencés par l’environnement. Aléatoires ou non, les facteurs de distorsion de ségrégation ont des effets considérables sur l’amélioration des cultures et la cartographie de leur génome. L’analyse de nombreux résultats impliquant une distorsion de ségrégation d’allèles a permis la caractérisation partielle des causes et des effets de ce phénomène. Les résultats obtenus montrent que l’importance de la DS dépend du type de marqueurs moléculaires utilisés et de la nature des populations étudiées. Des recherches complémentaires sont nécessaires pour une meilleure identification des facteurs générateurs de distorsion de ségrégation et une plus précise évaluation de leurs effets. La réalisation de cartographies génétiques denses, associée à des analyses cytogénétiques, devraient permettre de rapides progrès. Exemplaires

In Northern Cameroon, yield has been declining due to unsuitable cropping practices such as sub-optimal planting dates, along with an absence in genetic gain. The aim of this study was to use a cropping system model (DSSAT CSM-CROPGRO-Cotton) to identify the best cultivars (ideotypes) for Northern Cameroon that are adapted to low input rainfed productions systems for 2050 under RCP4.5 and RCP8.5. Calibration and evaluation of the CSM-CROPGRO-Cotton were performed with field observations for two cultivars (Allen Commun and L484). For RCP4.5 and RCP8.5, 50 replications for 2050 were generated based on an ensemble of 17 Global Circulating Models. In total, 3125 virtual cultivars representing existing genetic variability for phenology, morphology and photosynthesis were simulated. Thereafter, they were evaluated for performance under the projected future climate based on potential yield and the resilience of yield to suboptimal planting date. The widely cultivated cultivar L484 will be unsuitable under projected future climate, due to boll opening during the middle of the rainy season (median: 10/09 under RCP4.5 and 12/09 under RCP8.5). None of the ideotypes tested could optimize both yield and resilience (Pearson correlation<−0.82). However, compared to the current cultivar L484, two virtual ideotypes were identified: (a) “Ideo_sub” had a wide planting window, especially in the 10 worst replications of 2050, up to +5 days in RCP8.5; (b) “Ideo_Pot” had a high potential yield trait with low resilience to sub-optimal planting date, in the 10 worst replications of 2050, +530 kg ha−1 in RCP4.5 and +591 kg ha−1 in RCP8.5. Both ideotypes had an earlier anthesis date, a longer reproductive duration, and increase in the maximum photosynthetic rate. Therefore, breeding programs should consider these traits suggested by this system analysis using a crop simulation model for the identification of suitable cultivars under the projected future climate. The objective of this work was to evaluate shrub effect on soil nematode communities. Soil samples were collected from an experimental design where pearl millet (Pennisetum glaucum) was cultivated alone or with P. reticulatum stands and mulch. Soil nematofauna characteristics were determined and compared with results from soil under pure shrub stands and from bare soil. The analysis of soil nematofauna, characterized by the abundance of different trophic groups and related indices (MI, maturity index; EI and SI, enrichment and structure indices), allowed discrimination between treatments with or without shrub presence. The soil nematode community in millet cultivation was dominated by plant feeding nematodes, mainly from the Hoplolaimidae family, but their abundance decreased when P. reticulatum was associated to the cereal. The shrub also impacted other nematode trophic groups. The abundance of opportunistic bacterial feeders (mainly Cephalobidae) was increased in shrub treatments. Furtherresearch should explore consequences on cereal nutrition and nematicidal properties of P. reticulatum. Exemplaires

This study assessed the effect of two representative low and high quality residues (Zea mays and Crotalaria retusa respectively) on the formation and stability of soil aggregates. The formation and stability of aggregates, soil biomass, root biomass, fungal hyphae length, C mineralization and chitinase activity, as a specific biomarker of the activity of fungal populations, were measured under controlled conditions over 120 days. In both the
control and amended soils, there were more macroaggregates (>2000 mm) and mesoaggregates (250 e2000 mm) than microaggregates (50e250 mm and <50 mm). The formation of macroaggregates and stability (MWD) were not significantly affected by the quality of residues. Amendment with organic residues shifted the distribution of the aggregate fractions. The macroaggregates increased by 26% with crotalaria and by 35% with maize residues while mesoaggregates decreased by 18% with crotalaria and by 26% with maize residues and microaggregates decreased by 8% with crotalaria and by 9% with maize residues. This study also onfirmed that macroaggregates are formed from micro- and mesoaggregates. The total microbial biomass was significantly higher in soil amended with maize residues compared to oil with crotalaria residues and the control soil although the fungal hyphae length decreased when the soil was amended with either crop residue. Chitinase activity is the most pertinent indicator associated with acroaggregation stability. Adding mineral N (equivalent to 120 kg N ha1 as urea) to the residue increased microbial biomass and reduced fungal hyphae length but had no effect on macroaggregate formation and fungal activity. These observations suggested that, for short term incubation of soil amended with residues, fungal activity plays a greater role in aggregation in sandy soils than the fungal population density.Exemplaires

In order to evaluate the impact of genetic improvement on this decline, we conducted field experiments in two locations with 10 widely grown cotton cultivars released in Cameroon between 1950 and 2009. The rate of genetic gain (GG) was estimated with a linear regression of the cultivar mean on its year of release (YR). Contrasts between rates of GG observed with different planting dates were estimated and tested. Our results revealed a rate of GG on fibre yield of 3.3 kg ha−1 year−1 due to increased ginning out-turn (3.9% and 6.2% in 60 years in Garoua and Maroua, respectively). There was no GG on leaf area index (LAI), radiation use efficiency (RUE), aerial biomass, harvest index and on seed cotton yield. We concluded that cotton breeding efforts in Cameroon have successfully improved cotton fibre yield but there is still some room for seed cotton yield improvement. Exemplaires

The CROPGRO-cotton model was calibrated and evaluated at two locations in Cameroon over a period of two years using two planting dates and four contrasted cultivars. The model was used to assess yield gains by modifying plant traits such as specific leaf area, photosynthetic capacity and crop phenology. The ideotype was tested in conventional and conservation agriculture systems and under baseline and future climate conditions. The results revealed that, compared to existing cultivars, the ideotype requires longer to reach maturity and has thicker leaves with good photosynthetic capacity. In 2050 in North Cameroon, climate change will shorten time to maturity and cause a shift in the rainy season but neither change will have an effect on yields. Simulations with an ensemble of climate models revealed that models that assume higher rainfall predicted lower yields, suggesting that N leaching is a more important constraint than drought in North Cameroon. Our results will help cotton breeders select promising new traits to introduce in their cultivars for adaptation to climate changes in Cameroon and to similar sub-Saharan soil, cropping systems and climatic conditions.
