Genomic analysis of three African strains of Bacillus anthracis demonstrates that they are part of the clonal expansion of an exclusively pathogenic bacterium [texte imprimé] / L. Rouli, Auteur ; M. Symptomologie et dégats | Résumé : | Bacillus anthracis is the causative agent of anthrax and is classified as a ‘Category A’ biological weapon. Six complete genomes of B. anthracis
(A0248, Ames, Ames Ancestor, CDC684, H0491, and Sterne) are currently available. In this report, we add three African strain genomes:
Sen2Col2, Sen3 and Gmb1. To study the pan-genome of B. anthracis, we used bioinformatics tools, such as Cluster of Orthologous Groups,
and performed phylogenetic analysis. We found that the three African strains contained the pX01 and pX02 plasmids, the nonsense
mutation in the plcR gene and the four known prophages. These strains are most similar to the CDC684 strain and belong to the A cluster.
We estimated that the B. anthracis pan-genome has 2893 core genes (99% of the genome size) and 85 accessory genes. We validated the
hypothesis that B. anthracis has a closed pan-genome and found that the three African strains carry the two plasmids associated with
bacterial virulence. The pan-genome nature of B. anthracis confirms its lack of exchange (similar to Clostridium tetani) and supports its
exclusively pathogenic role, despite its survival in the environment. Moreover, thanks to the study of the core content single nucleotide
polymorphisms, we can see that our three African strains diverged very recently from the other B. anthracis strains. |

en réponse à l’inoculation mycorhizienne arbusculaire. Les essais conduits en serre pendant trois mois, ont
montré un meilleur développement et une meilleure nutrition minérale chez les plants inoculés comparé aux
témoins non inoculés. Les variétés testées se sont révélées très dépendantes de la mycorhization arbusculaire. Le
degré de dépendance est généralement supérieur à 55 % et est fortement corrélé à l’isolat fongique et à la variété
testée. Il ressort de cette étude que les biofertilisants à base de champignons mycorhiziens procurent des
bénéfices importants à la plante, et peuvent être utilisés pour améliorer la croissance et le rendement du sésame |

There were six available complete genomes (A0248, Ames, Ames Ancestor, CDC684, H0491 and Sterne). Here, one Gambian and two Senegalese strains (Gmb1, Sen2Col2 and Sen3) were added. In this work, the pan-genome of B. anthracis was studied based on nine strains and using bioinformatics tools as Cluster of Orthologous Groups (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG). Thereafter, B. anthracis pan-genome having 2893 core genes and 85 accessory genes was estimated. With Mauve method, the pan-genome of B. anthracis was verified and it was found to be very narrow and clonal. To have confidence in this study, different tools were used to compare and validate the results. All of the tools yielded the same results; the addition of the Senegalese and Gambian strains did not change the nature of the B. anthracis pan-genome (2893 core genes and 85 accessory genes), which had a core/pan-genome ratio of 99%. The closed nature of the pan-genome of B. anthracis (the core genome) represents 99% of the pan-genome size. The hypothesis that B. anthracis had a closed pan-genome was hereby validated. |

Six génomes complets de Bacillus anthracis (AO248,Ames, Ames Ancestor, CDC 684, H9401 et Sterne) ont été rajoutés aux trois génomes Sénégalais (Sen1, Sen2, Sen3) et Gambien (Gmb1).
Nos travaux sont axés sur la caractérisation des séquences génomiques de Bacillus anthracis pour les souches sénégalaises et gambienne par comparaison à d’autres souches de référence pour préciser la nature exacte du pan-génome. Résultats : Nous avons trouvé que les souches sénégalaises et gambienne étaient porteuses des deux plasmides pXO1 et pXO2. Les séquences oligonucléotidiques de ces souches montrent une identité de séquence dans les deux pays ciblés et le pan-génome a été estimé comme renfermant 2893 cores génomes,7 gènes uniques et 85 gènes accessoires.
Cette séquence de Bacillus anthracis a été comparée et est identique à celle obtenue à partir du génome de la souche Sterne de la souche CDC 684 et de celle de la H9401 trouvée chez un humain atteint d’Anthrax en Corée. Nous avons validé l’hypothèse selon laquelle les souches de Bacillus anthracis testées ont une pan-génome de type fermé.
